The key parameters that should be considered during formulation development include:

  • The physicochemical properties of the active ingredient(s) – solubility, particle size, pKa, stability.
  • The route of administration – oral, topical, injectable etc.
  • The desired release profile – immediate release, controlled release, sustained release etc.
  • The target patient population – pediatric, geriatric etc.
  • Other excipients that may affect the performance of the final product – fillers/binders/diluents, surfactants, emulsifiers etc.

Current Perspective

The current perspective on formulation and development is that it should be approached from a systems perspective. This means taking into account the interactions between the different components of the system (e.g., drug, excipients, packaging) as well as the environment (e.g., storage, use conditions). In addition, formulation and development should be considered as an iterative process, with each iteration bringing the product closer to its final form.
